Since there is no
universal convention about what values trigger the requirement for an
additional signature, a bank would not refuse to honor a check with
only one signature where two are required by the check issuer, unless
the issuer makes this requirement explicit.  

Precisely.  The issuer makes this requirement explicit by placing it into the
content, and the context (namely, banking policy that negotiable
instruments may contain arbitrary restrictions as part of their content).
The only standardized part of a check is the MICR encoding on the bottom.
Not coincidentally, this is a representational standard, not a semantic one...
