I want to cover any number of spaces before the word 'SPAM:' (should I  
escape the : with a \?)

I usually use .* but I don't want *any* character, I just want any number of  
_spaces_

Why don't you use the following (it also matches 'tabs'):

* ^Subject:[    ]*SPAM:

(Note that the items in between '[]' are a blank and a hard tab.
