Need more info.
Try "procmail -v" and see which locking strategies are
being used... does your system support the locking strategies listed?
Also it is possible if a second (or more) instance of mail is
reading mail from the spool file, there is a legitimate lock on
the file.
Is this a new installation of procmail? Did you follow
the instructions in the INSTALL/README regarding
configuration?
