You mean by using a header field "Type: mailing-list-subscription-request" or
something like that? And that spam traps should ignore messages with that
header?
That won't work unless the protocol dictates that such requests shall never
have to be seen/read by a human, but shall be interpreted and answered by the
user's MUA automatically. Otherwise, spammers would just (falsely) use that
header, too.
